DNA-Mismatch Repair: The intricacies of eukaryotic spell-checking
نویسنده
چکیده
Recent work suggests that the eukaryotic system responsible for repairing DNA mismatches, and so correcting replication errors, is more complex than was thought; its multiple components have many cellular functions.

Biochemistry and genetics of eukaryotic mismatch repair.
The process of mismatch repair was first postulated to explain the results of experiments on genetic recombination and bacterial mutagenesis. Mismatch repair has long been known to play a major role in two cellular processes: (1) the repair of errors made during DNA replication or as the result of some types of chemical damage to DNA and DNA precursors; and (2) the processing of recombination i...
متن کاملDNA mismatch repair in plants. An Arabidopsis thaliana gene that predicts a protein belonging to the MSH2 subfamily of eukaryotic MutS homologs.
Sets of degenerate oligomers corresponding to highly conserved domains of MutS-homolog (MSH) mismatch-repair proteins primed polymerase chain reaction amplification of two Arabidopsis thaliana DNA fragments that are homologous to eukaryotic MSH-like genes. Phylogenetic analysis places one complete gene, designated atMSH2, in the evolutionarily distinct MSH2 subfamily.
متن کاملA possible mechanism for exonuclease 1-independent eukaryotic mismatch repair.
Mismatch repair contributes to genetic stability, and inactivation of the mammalian pathway leads to tumor development. Mismatch correction occurs by an excision-repair mechanism and has been shown to depend on the 5' to 3' hydrolytic activity exonuclease 1 (Exo1) in eukaryotic cells. However, genetic and biochemical studies have indicated that one or more Exo1-independent modes of mismatch rep...
متن کاملThe Eukaryotic Mismatch Recognition Complexes Track with the Replisome during DNA Synthesis
During replication, mismatch repair proteins recognize and repair mispaired bases that escape the proofreading activity of DNA polymerase. In this work, we tested the model that the eukaryotic mismatch recognition complex tracks with the advancing replisome. Using yeast, we examined the dynamics during replication of the leading strand polymerase Polε using Pol2 and the eukaryotic mismatch reco...
متن کاملMitochondrial DNA repair and damage tolerance.
The accurate maintenance of mitochondrial DNA (mtDNA) is required in order for eukaryotic cells to assemble a functional electron transport chain. This independently-maintained genome relies on nuclear-encoded proteins that are imported into the mitochondria to carry out replication and repair processes. Decades of research has made clear that mitochondria employ robust and varied mtDNA repair ...
